  • Sucrose 103.00 g
  • K2SO4 0.25 g
  • MgCl2.6H2O 10.12 g
  • Glucose 10.00 g
  • Casaminoacids 0.10 g
  • Trace element solution 2 ml
  • Yeast extract 5.00 g
  • TES buffer 5.73 g
  • Distilled water up to 1 L

Pour 100 ml of the solution into 250 ml Erlenmeyer flasks each containing 2.2 g agar. Close the flasks and autoclave. At time of use, re-melt the medium and supplement with the following ingredients in the order listed:

  • KH2PO4 (0.5%) 1 ml
  • CaCl2.2H2O (5 M) 0.4 ml
  • L-proline (20%) 1.5 ml
  • NaOH (1 N) 0.7 ml
